Following ligand stimulation, receptor tyrosine kinases are internalised in the endosome, where they undergo limited dephosphorylation by PTP1B present on the endoplasmic reticulum.

Cell Sci. 114:2001;2167-2178 This article provides convincing evidence that Cbl-mediated ubiquitination of epidermal growth factor receptors (EGFRs) occurs at the plasma membrane, a topic that has been debated for a long time. • The endophilin-CIN85-Cbl complex mediates ligand-dependent downregulation of c-Met
    • ••]) identified a novel function of Cbl in ligand-induced downregulation of receptor tyrosine kinases. Cbl was shown to interact with the CIN85-endophilin complex thus controlling internalisation of epidermal growth factor and c-Met receptors. This pathway was functionally separable from the ubiquitin ligase activity of Cbl and was dependent on direct binding of CIN85 to the evolutionary divergent carboxyl terminus of Cbl.

    • A single motif responsible for ubiquitin recognition and monoubiquitination in endocytic proteins

UIMs are found usually in endocytic proteins (e.g. Eps15R related, epsins and Hrs) that are also monoubiquitinated after ligand stimulation. The existence of a UIM/ubiquitin-based network involved in regulation of different steps in the endocytic pathway is proposed.

The conserved carboxy-terminal domain can inhibit EGF-induced mitogen-activated protein kinase (MAPK) activation, while the amino-terminal part of Sprouty2 sequesters Cbl and inhibits EGFR ubiquitination and degradation, thus potentiating EGFR signalling via the MAPK pathway.
