(2000) [12•] demonstrate that Hrd1p is the rate-limiting ubiquitin ligase for degradation of Hmg2p (and other ER proteins) in yeast, and that Hrd1p, with Hrd3p, forms a transmembrane ubiquitin ligase that functions on both sides of the membrane. This study identifies an ER-anchored ubiquitin ligase, Doa10p, which is completely distinct from Hrd1p/Der3p; demonstrating that there are multiple ER-associated degradation pathways working together to manage unfolded ER proteins.
