    • This study demonstrates the importance of membrane-type MMPs, rather than soluble MMPs, in cell migration and morphogenesis. Therefore, membrane-type MMPs appear to be better candidates than soluble MMPs as targets for antiangiogenic therapy.

    • This is the first systematic analysis of gene expression in tumor endothelium. Gene expression profiles of highly purified endothelial cells from human colorectal carcinomas and endothelial cells from normal tissues were compared using serial analysis of gene expression (SAGE). The analysis led to the identification of several panendothelial-cell markers (expressed on all endothelial cells, but not on other cell types) and of genes differentially expressed on endothelial cells derived from tumor versus normal tissues (genes upregulated in tumor endothelium or with higher expression in normal endothelium). Many of these genes still need to be characterized.

    • Different schedules of treatment with four antiangiogenic compounds were investigated in a transgenic mouse model of pancreatic islet multistage carcinogenesis (RIP1-Tag2). The MMP inhibitor BB-94 and endostatin were most effective when treatment was given at the hyperplastic stage, to block the angiogenic switch, or on small, asymptomatic tumors. They were not effective in mice with a substantial tumor burden. AGM-1470 (TNP-470, see Table 1) did not prevent the angiogenic switch, but was effective on mice bearing small, vascularized tumors. Interestingly, only treatment with AGM-1470 and with the combination of endostatin and angiostatin caused a regression of large tumors, although all four compounds were effective in increasing the survival of mice.
