TNFα-induced secretion of RANTES and IL-6 from human airway smooth muscle cells: Modulation by cAMP
-
This paper demonstrates that elevation of cAMP within human ASM cells augments TNF-induced IL-6 secretion, but inhibits TNF-induced RANTES secretion. These effects are independent of cytokine-induced activation of NF-κB. Pharmacologic agents that modulate levels of cAMP may therefore have different effects on airway inflammation.

Sphingosine 1-phosphate modulates human airway smooth muscle cell functions that promote inflammation and airway remodeling in asthma

10.1096/fj.00-0742fje. This paper describes the effects of sphingosine-1-phosphate (SPP) on a range of smooth-muscle cell functions including calcium mobilization, proliferation and cytokine secretion. It is also the first description of SPP in the airways of asthmatics following antigen challenge and suggests that SPP may be an important mediator of airway inflammation

Constitutive expressions of type I NOS in human airway smooth muscle cells: Evidence for an antiproliferative role
-
Exhaled nitric oxide (NO) levels are increased in the airways of asthmatics and is believed to be a marker of airway inflammation. In this paper, the authors demonstrate that human ASM cells express NO synthase, which results in an inhibition of smooth muscle cell proliferation. This suggests an autocrine feedback loop that may have a protective effect on airway remodeling.

Up-regulation of ICAM-1 by cytokines in human tracheal smooth muscle cells involves an NF-κB-dependent signaling pathway that is only partially sensitive to dexamethasone
-
The authors demonstrate that several of the pro-inflammatory effects of TNF on ASM cells occur through steroid-independent pathways. These data are important, as steroids are a mainstay of therapy in asthma; however, not all inflammatory responses are modulated by these agents.
