Mice lacking pulmonary surfactant protein-A were infected with respiratory syncytial virus by intratracheal instillation. Pulmonary infiltration was more severe in the SPA -/-mice compared with controls, and was associated with an increased number of RSV plaque-forming units in lung homogenates. Concentrations of TNFα and IL-6 were increased in the SP-A knock-out mice; they were noted to have decreased superoxide and hydrogen peroxide generation. Co-administration of exogenous SP-A reduced viral titers and inflammatory cells in the SP-A -/-mice. These findings demonstrate the host defense rote of SP-A in vivo.

SP-A -/-mice were infected by intratracheal injection with replication-deficient recombinant adenovirus containing a luciferase marker gene. Lung inflammation was markedly increased in the SP-A -/-mice compared with wild-type mice and was associated with increased hemorrhage and epithelial cell injury. Co-administration of purified human SP-A reduced adenoviral-induced lung inflammation in the SP-A -/-mice. This study shows that SP-A enhances viral clearance and inhibits lung inflammation during pulmonary viral infection.

J Immunol 1999, 163:4495-4502. The effect of recombinant human SP-A and natural canine SP-A was studied on the human alveolar macrophage response to Candida albicans. Both forms of SP-A reduced the expression of TNFα, IL-1β, Mip-1α, and MCP-1 in a dose-dependent manner. SP-A did not affect baseline liberation of these cytokines. Release of the anti-inflammatory cytokines IL-1 receptor antagonist and IL-6 was not inhibited by SP-A. The effect of SP-A required internalization by the mononuclear phagocytes, suggesting that it has an effect on the expression of pro-inflammatory cytokine synthesis at the transcriptional level.

Eur J Immunol 1999, 29:3478-3484. The effects of recombinant trimeric SPD lectin domains was studied in relation to the binding of RSV-G protein. The study was performed by infection of monolayers of HEP-2C cells and in vivo infections in BALB/c mice. Full-length and recombinant SP-D bound to RSV-G protein in a concentration-dependent manner. Binding was inhibited by EDTA and mannan, indicating that binding occurs via carbohydrate recognition domain of SP-D. Recombinant SPD inhibited RSV infectivity in cell culture in a dose-dependent manner. Intranasal administration of recombinant SPD to RSV-infected mice inhibited replication in the lungs and reduced levels of virus by 80%.

SP-D bound to live P. aeruginosa, and in vitro killing assays demonstrated that macrophage-mediated killing of one of six strains of P. aeruginosa was modestly enhanced by SP-D.

J Immunol 1999, 163:312-321. These authors demonstrated that specific binding of SP-D to M. tuberculosis was saturable, calcium dependent, and inhibitable by carbohydrates. Incubation of M. tuberculosis with SP-D led to agglutination of the bacteria. Incubation of M. tuberculosis with subagglutinating concentrations of SP-D led to reduced adherence of the bacteria to macrophages, whereas adherence was increased by incubation with SP-A.

This important study showed the human beta-defensins are chemotatic for immature dendritic cells and memory T cells. When these cells were stably transfected with human chemokine receptor 6 (CCR6), which they usually express, they demonstrated selective chemotaxis to human beta-defensin. This induced chemotaxis was sensitive to pertussis toxin and inhibited by antibodies to CCR6. The binding of a labeled ligand to CCR6 was competitively displaced by beta-defensin. These results suggest that β-defensins may facilitate specific immune response by recruiting dendritic and T cells to the sites of infection by interacting with CCR6.

Levels of the peptide were increased three-to fourfold over baseline by this treatment, and bacterial killing activity was restored to normal levels.

To investigate overexpression of the antimicrobial peptide LL-37/hCPA-18 in the lungs, C57BI/6 mice received an adenoviral vector containing the cDNA for this peptide. Mice treated with the vector had a lower bacterial load and a reduced inflammatory response after pulmonary challenge with P. aeruginosa. Systemic expression of the vector by intravenous injection resulted in improved survival rates following intravenous injection of E. coli or lipopolysaccharide. This important study demonstrates that expression of an antimicrobial peptide by gene transfer results in augmentation of the innate immune response.

CCSP knock-out mice were exposed to a recombinant E1-and E3-deleted adenoviral vector containing the luciferase marker gene. Lung inflammation was markedly increased in the CCSP knock-out mice compared with wild-type controls and associated with an increased number of polymorphonuclear cell infiltrates and increased epithelial cell injury. This increased injury was associated with increased concentrations of IL-1β, TNFα, Mip-1α, and MCP-1. Adenoviral-mediated gene transfer was decreased in the knock-out mice compared with wild-type controls. This study indicates that CCSP modulates lung inflammation in response to viral infection.

This paper reviews the activities of lactoferrin. Lactoferrin modulates the inflammatory response by inhibiting release of cytokines from monocytes and regulates in the proliferation differentiation of immune cells. Some of these activities are related to the ability of lactoferrin to bind LPS with high affinity and to prevent its binding to CD14.

The first six residues of the N-terminus were critical for antiendotoxin activity. Co-injection of LF-33 with E. coli LPS significantly reduced the lethality of LPS in the galactosamine-sensitized mouse model. This peptide may have a future role for the treatment of endotoxin-induced septic shock.

The CD28/B7 interaction provides essential costimulatory signals for T cell activation and differentiation. Alveolar macrophages from patients with hypersensitivity pneumonitis have upregulated expression of B7 molecules. The authors investigated the activity of CTLA4-Ig, a CD28/B7 antagonist, on lung inflammation in mice inoculated with Saccharopolyspora rectivirgula, compared with the group that received a control Ig. The mice that received CTLA4-Ig had markedly decreased lung inflammation, and production of proinflammatory cytokines was also increased. The study shows that blockade of CD28/B7 interactions by CTLA4-Ig may provide a novel approach to treatment of hypersensitivity pneumonitis.
