Siderophore-mediated iron transport: Crystal structure of FhuA with bound lipopolysaccharide
-
FhuA, the receptor for ferrichrome-iron in Escherichia coli, is a member of the TonB-dependent outer membrane receptors. Its three-dimensional structure has been determined with and without ferrichrome-iron. FhuA is a β-barrel composed of 22 antiparallel β-strands that is closed in the periplasm by the amino-proximal domain named the cork. Upon binding of ferrichrome-iron, conformational changes are transduced to the periplasmic pocket of FhuA, signaling the ligand-loaded status of the receptor

Extracellular haemophores are found in Serratia marcescens, Pseudomonas aeruginosa and Yersinia pestis. Both haem-free (apoprotein) and haem-loaded (holoprotein) hemophores bind to receptors demonstrating direct protein-protein interactions, which is TonB independent
