Key findings are that the effects of MCP-1 may be confined to specific compartments within the kidney, that complete absence of MCP-1 is associated with significant but not complete protection from immune injury, and that MCP-1 deficiency may affect expression of other cytokines and chemokines.

This model displays elevated urinary MCP-1 levels, proteinuria, and interstitial monocyte infiltration, which can be reversed with angiotensin converting enzyme inhibition. These deleterious effects are not due to hypertension alone.

This unexpected outcome may be because the absence of CCR1 appears to favor a Th1 immune response. This provocative study adds another level of complexity to the issue of manipulating the chemokine network for therapeutic benefit.

The role of angiotensin II in anti-GBM glomerulonephritis is explored by inducing nephritis in the angiotensin II type 1a receptor knockout mouse. Relevant to chemokine regulation, this receptor knockout has no effect on early (6 h) MCP-1 expression, but prevents the rise in MCP-1 after 1-2 weeks. The renin-angiotensin system is activated in this model at day 1, after which activity declines. It is interesting that the effects of angiotensin II are delayed until a more chronic phase of disease.
