This is the first application of random synthetic peptide libraries for the definition of human MHC class 1 binding motifs (HLA-A2) using MHC-peptide complex refolding. The motif defined by elution could be confirmed. This study demonstrated that this approach is successful providing that MHC-peptide complexes are void of nonspecifically binding peptides.

Definition of natural T cell antigens with mimicry epitopes obtained from dedicated synthetic peptide libraries

Using the technology described in [23,24], mimicry epitopes derived from one-bead-one-peptide libraries are found for three different clones. In all cases, these mimicry epitopes showed high similarity to their natural counterparts. This indicates that synthetic libraries can now be used generally for T cell antigen definition.
