Cell 1999, 97:41-51. This paper describes the purification of a four-subunit, negative elongation factor complex that binds tightly to RNAPII and is critical for DRB-sensitive transcription. NELF is shown to act co-operatively with DSIF and the NELF-E subunit is identified as RD, a protein containing an RNA-recognition motif.

Interestingly, the ability of FCP1 to act as an ELF does not require its CTD phosphatase activity.

    • Garber M, Wei P, Kewal-Ramani V, Mayall T, Herrmann C, Rice A, Littman D, Jones K: The interaction between HIV-1 Tat and human cyclin T1 requires zinc and a critical cysteine residue that is not conserved in the murine CycT1 protein. Genes Dev 1998, 12:3512-3527. This paper demonstrates that Cys261 in the CycT1 TRM is necessary for binding to Tat and TAR RNA in vitro and that zinc is required for the protein-protein interaction. The murine CycT1 is shown to contain a tyrosine at position 261 and to be unable to support Tat Trans-activation in vivo; these defects are repaired by a point mutation that restores Cys261. Stable rodent cell lines expressing human CycT1 are shown to support transcription from an integrated HIV-1 provirus but to be unable to support virus replication. The defect in the mouse CycT1 protein was also reported elsewhere [26-28,31].
