The murine gammaherpesvirus 68 v-cyclin is an oncogene that promotes cell cycle progression in primary lymphocytes
-
Van Dyk LF, Hess JL, Katz JD, Jacoby M, Speck SH, Virgin HW: The murine gammaherpesvirus 68 v-cyclin is an oncogene that promotes cell cycle progression in primary lymphocytes. J Virol 1999, 73:5110-5112. The authors showed that open reading frame (orf) 72 of γHV68 is a bona fide gene encoding v-Cyclin. Transgenic studies showed that v-Cyclin promotes cell cycle progression in thymocytes and induces lymphoblastic lymphoma in a significant fraction of transgenic mice. This identifies v-Cyclin as the first γHV68 oncogene.

Analysis of murine gammaherpesvirus-68 transcription during lytic and latent infection
-
Simas JP, Swann D, Bowden R, Efstathiou S: Analysis of murine gammaherpesvirus-68 transcription during lytic and latent infection. J Gen Virol 1999, 80:75-82. The authors perform an extensive transcriptional analysis in vitro and in vivo and find that the M3 genome region of γHV68 is transcribed in the spleen at times when many other probes are negative. This suggests an interesting role for M3 in γHV68 infection, perhaps during the transition to latency.

B cells regulate murine gammaherpesvirus 68 latency
-
Weck KE, Kim SS, Virgin HW IV, Speck SH: B cells regulate murine gammaherpesvirus 68 latency. J Virol 1999, 73:4561-4661. The authors demonstrate that B cell deficient mice maintain a high frequency of cells that reactivate latent γHV68 at times after infection when normal mice are transitioning from having a high frequency of cells that reactivate the virus to a low frequency. This paper presents the first assay for quantitating the frequency of cells carrying γHV68 genome during latency. The authors use this assay to demonstrate that B cells regulate the efficiency of reactivation from latency rather than the frequency of cells carrying γHV68 genome.

Macrophages are the major reservoir of latent murine gammaherpesvirus 68 in peritoneal cells
-
Weck KE, Kim SS, Virgin HW IV, Speck SH: Macrophages are the major reservoir of latent murine gammaherpesvirus 68 in peritoneal cells. J Virol 1999, 73:3273-3283. The authors show that macrophages carry latent γHV68 and confirm that B cells carry latent γHV68 genome during latency. This is an important finding as it shows that there are multiple cellular reservoirs of latent γHV68. In addition they show that latent cells can be recruited into sites of inflammation, arguing that a reservoir of cells in the bone marrow may supply latent cells to tissues.

Lung epithelial cells are a major site of murine gammaherpesvirus persistence
-
Stewart JP, Usherwood EJ, Ross A, Dyson H, Nash T: Lung epithelial cells are a major site of murine gammaherpesvirus persistence. J Exp Med 1998, 187:1941-1951. The authors present data clearly showing lytic infection in the lung of B cell deficient mice at late times after infection. They provide data for a role for B cells in spreading γHV68 and argue that lung epithelial cells are latently infected with γHV68.

Three distinct regions of the murine gammaherpesvirus 68 genome are transcriptionally active in latently infected mice
-
Virgin HW, Presti RM, Li X-Y, Liu C, Speck SH: Three distinct regions of the murine gammaherpesvirus 68 genome are transcriptionally active in latently infected mice. J Virol 1999, 73:2321-2332. This is the first paper to clearly demonstrate transcription of the γHV68 genome in latent tissues. Several candidate latency genes are identified. Interestingly, several of these are shared with primate γ-herpesviruses EBV, HVS and KSHV. This argues that γHV68 latency will share important molecular mechanisms of latency with primate γ-herpesviruses.

Non-antigen-specific B-cell activation following murine gammaherpesvirus infection is CD4 independent in vitro but CD4 dependent in vivo
-
Stevenson PG, Doherty PC: Non-antigen-specific B-cell activation following murine gammaherpesvirus infection is CD4 independent in vitro but CD4 dependent in vivo. J Virol 1999, 73:1075-1079. The authors show that in vitro infection of murine B cells results in striking activation of B cells. Given the role of B cells in multiple aspects of γHV68 infection, this is probably an important mechanism for γHV68 pathogenesis.
