A histone H2B-GFP fusion protein was made and characterized. HeLa cells stably expressing the fusion protein showed no change in cell cycle parameters. In vivo imaging using this fusion protein to visualize the intranuclear chromatin distribution revealed fine substructure consistent with the large-scale chromatin organization visualized previously by DAPI (4,6-diamino-2-phenylindole) staining and deconvolution microscopy in aldehyde fixed cells.

Microinjection of fluorescent nucleotides into mammalian cells followed by long chases of several cell cycles and random chromosome segregation produced cells containing a small number of labeled chromosome regions. This system then allowed in vivo visualization of the dynamics of native chromosome domains, confirming the confinement of chromosome regions to compact territories. The shape and position of these regions were stable in general over several hours but there were examples in which small changes in shape and position were visualized.

A highly reproducible pattern of condensation and decondensation tightly coupled to cell cycle progression was observed in vivo for a specific amplified chromosome region. Significant large-scale chromatin decondensation appeared to slightly precede initiation of DNA replication and movement of the amplified chromosome region into the nuclear interior.

Simulations of the dynamics of this model were consistent with previous FISH experimental observations of distinct chromosome territories. Simulations were also consistent with published FISH data measuring the distances between pairs of probes, which had previously been interpreted in terms of a giant-loop model.

MIX-1, belonging to the SMC protein family, is shown to be a component of the dosage compensation complex that associates with X chromosomes to reduce gene expression in addition to being a component of the mitotic SMC complex associating with mitotic chromosomes. Mutations in this gene disrupt both dosage compensation and mitosis. Assuming that the dosage compensation complex regulates chromosome condensation similarly to the mitotic SMC complex, this work provides perhaps the strongest evidence to date linking large-scale chromatin structure and transcriptional regulation.

If large-scale chromatin organization plays a functional role in regulating gene activity, then one would expect chromosome regions rich in housekeeping genes to show a different structure to that of gene poor regions. FISH was used to measure the interphase distances between pairs of probes separated by varying genomic distances within 2 G-band and 3 R-band chromosome regions. Distributions of measured interphase distances indicated a more open chromosome structure for R band regions.

In differentiated ES cells and erythroleukemia cells containing this chromosome, a global increased sensitivity to DNasel and developmental transcriptional regulation of the locus was maintained; however, transcription of β-globin was reduced to 5-25% of normal levels. These results suggest that additional sequences, further upstream, are capable of establishing the locus open chromatin state.

    • The locus control region is necessary for gene expression in the human beta-globin locus but not the maintenance of an open chromatin structure in erythroid cells
    • 1 the human β-globin locus LCR was deleted at the native chromosome location using homologous recombination in tissue culture cells and transferred to mouse erythroleukemia cells. Although transcription of all β-globin genes was eliminated, an open chromatin state marked by global increased DNAsel sensitivity remained. Therefore, the LCR as originally defined is not necessary to create the open chromatin state. Also, this work shows the open chromatin state is not sufficient by itself to allow transcription.

Transgene arrays with insulators produced more consistent expression levels at different chromosome sites and were protected from gradual extinction of transcription during long-term culture. Changes in nuclease accessibility, DNA methylation and histone acetylation accompanied this extinction with uninsulated arrays but changes in nuclease accessibility and histone acetylation were prevented with insulators. Extinction of expression occurred in an all or nothing response over the entire transgene arrays.

The predominant component of this heterochromatin is satellite 3, containing SAR activity, making a giant, reiterated SAR. MATH20 expression suppressed topoisomerase 2 binding to this satellite repeat. The authors propose that MATH20 displaces a heterochromatin protein(s), which might be related to SAR function.
