The authors showed that modifications to glucoamylase, which had increased selectivity to maltose versus isomaltose as a substrate, could yield higher peak dextrose levels in commercial modeled saccharification reactions.

Li Y, Reilly PJ, Ford C: Effect of introducing proline residues on the stability of Aspergillus awamori. Protein Eng 1997, 10:1199-1204. The authors show that proline substitutions can affect stability against denaturation, which may or may not be also seen in stability to thermoinactivation. Interestingly, one specific substitution, Ser460→Pro showed increased thermoinactivation, presumably due to removal of an O-glycosylation site, showing the importance of post-translational modification in functionality of glucoamylase.

A thermostable pullulanase is described with a temperature optimum above 100°C and a pH optimum of 5.5, which is consistent with low pH amylases and opens the possibility for a single liquefaction/debranching step for starch production.
