The authors performed an analysis of specific lysine residues in the amino-terminal tail of histone H4 and showed that, in the absence of the histone H3 tails, mutations of K5 and K12 to glycine residues had no effect on cell viability or nucleosome assembly whereas mutations in all three lysine residues, (K5, K8 and K12) were lethal.

The cloning of the human HAT1 cDNA and purification and characterization of the holoenzyme comprising the catalytic (HAT1) and regulatory (RbAp46) subunits from human 293 cell extracts are the focus of this paper. The authors show that RbAp46 could bind histone H4 in the absence of the catalytic subunit and that the holoenzyme acetylated free but not nucleosomal histone H4 and that HAT1 was predominantly nuclear in S-phase.

Genes Dev 1997, 11:345-357. The purification and cloning of yeast CAF1 and the characterization of phenotypes associated with null mutants in the CAF1 subunits are described in this report. The deletion of CAF1 subunits is not lethal in yeast suggesting that other pathways exist for the deposition of core histones onto replicated DNA.

Genes Dev 1997, 11:358-370. In this paper, the authors report the identification of CAC1 in a genetic screen for genes that alleviated antagonism between telomeres and centromeres. They also show that Rap1p was mislocalized in ca1 mutants which are also defective in silencing at telomeres.

They have also identified an interaction between Cac1p and the DNA helicase Pif1p that is thought to inhibit telomere replication.

The authors demonstrate that loss of the HIR genes (which regulate histone biosynthesis) in a CAF1 mutant background leads to a synergistic decrease in silencing at telomeres and at the HM loci. However, hir mutants alone had no silencing defects, suggesting that these proteins affect the alternative assembly pathway.

In a series of elegant experiments, the authors performed a microscopic analysis of cac1 mutants and show that loss of HML silencing was most likely caused by an inability to maintain the repressed chromatin structure, itself presumably a result of incomplete or inappropriate nucleosome assembly. The maintenance defect was alleviated by increased dosage of SIR3 but not SIR1.
