Conversely, overexpression of the bFGF gene driven by a rhodopsin promoter induced marked increases in photoreceptor bFGF expression without inducing retinal neovascularization or increasing retinal neovascularization under ischemic conditions. This is the most conclusive evidence to date that bFGF is neither necessary nor sufficient to induce retinal neovascularization.

Ischemic retinal neovascularization was inhibited in transgenic mice expressing a growth hormone (GH) antagonist gene and in normal mice given an inhibitor of GH secretion. Inhibition was inversely proportional to serum levels of GH and a downstream effector, IGF-I. Inhibition was reversed with exogenous IGF-I administration. GH inhibition did not diminish hypoxia-stimulated retinal VEGF or VEGF receptor expression. GH overexpression did not increase neovascularization. These data suggest that GH and IGF-1 are probably permissive factors for neovascularization and that systemic inhibition of GH or IGF-I, or both, may have therapeutic potential in preventing diabetic retinopathy.

Additional data suggest that Sp1 is necessary for KDR promoter activity in endothelial cells and is sufficient to induce KDR expression in nonendothelial cells, thus possibly accounting for cell-type expression specificity.
