Mutant Chinese hamster ovarian (CHO) cells that lack the Site-2 protease (M19 cells) were permanently transfected with a cDNA encoding either nuclear SREBP-1a, -Ic or 2. A muristerone-inducible promoter was used for the SREBP constructs that allowed for graded expression of each SREBP isoform. The M19 cells lack all endogenous nuclear SREBPs, owing to their inability to cleave the membrane-bound SREBP precursors. Therefore, the effect of each SREBP isoform on potential SREBP target genes could be determined. This is the best system currently available to test the individual roles of each SREBP isoform in cell culture.
