Cleavage of sterol regulatory element-binding proteins (SREBPs) at site-1 requires interaction with SREBP cleavage-activating protein. Evidence from in vivo competition studies

Sakai J, Nohturfft A, Goldstein JL, Brown MS. Cleavage of sterol regulatory element-binding proteins (SREBPs) at site-1 requires interaction with SREBP cleavage-activating protein. Evidence from in vivo competition studies. J Biol Chem 1998; 273:5785-5793 A series of elegant studies demonstrating the COOH-terminal domains of SREBP and SCAP must form a complex to direct Site-1 cleavage of SREBPs. The importance of this paper demonstrates that SCAP is not only a sterol-sensor, but also is physically required for cleavage of SREBPs at Site-1.

Molecular identification of the sterol-regulated luminal protease that cleaves SREBPs and controls lipid composition of animal cells. Mol Cell 1998, 2.505-514. This paper reports the cloning of the Site-1 protease (S1P) using an expression cloning strategy in cholesterol auxotrophic cells that lacked S1P The cloned cDNA encodes a 1052-amino acid membrane-bound subtilisin-like protease. The cloning of this protein is a crucial advance for the complete understanding of sterol-mediated regulation of SREBP cleavage

Differential stimulation of cholesterol and unsaturated fatty acid biosynthesis in cells expressing individual nuclear sterol regulatory element-binding proteins
-
Pai J-T, Guryev O, Brown MS, Goldstein JL. Differential stimulation of cholesterol and unsaturated fatty acid biosynthesis in cells expressing individual nuclear sterol regulatory element-binding proteins. J Biol Chem 1998, 273:26138-26148. Mutant Chinese hamster ovarian (CHO) cells that lack the Site-2 protease (M19 cells) were permanently transfected with a cDNA encoding either nuclear SREBP-1a, -Ic or 2. A muristerone-inducible promoter was used for the SREBP constructs that allowed for graded expression of each SREBP isoform. The M19 cells lack all endogenous nuclear SREBPs, owing to their inability to cleave the membrane-bound SREBP precursors. Therefore, the effect of each SREBP isoform on potential SREBP target genes could be determined. This is the best system currently available to test the individual roles of each SREBP isoform in cell culture.
