Hepatology 1998; 28:1637-1644. The impaired canalicular secretion associated with endotoxin-induced cholestasis was associated with a significant down-regulation of the mRNA and protein for the canalicular organic anion transporter, Mrp2, and a partial down-regulation of the BSEP. This was one of the first studies of BSEP regulation.

J Lipid Res 1999; 40:302-308. The percentage cholesterol absorption was quantified using dual stable isotopic tracers and negative ion mass spectrometry in normal women and men consuming a moderately low cholesterol containing diet. Cholesterol absorption values ranged widely from 29% to 80%, but were not related to plasma total or LDL cholesterol in these individuals.

U-shape relationship between change in dietary cholesterol absorption and plasma lipoprotein responsiveness and evidence for extreme interindividual variation in dietary cholesterol absorption in humans. J Lipid Res 1998; 39:2415-2422. The efficiency of cholesterol absorption was measured in human subjects on diets containing low fat/low cholesterol, high fat/low cholesterol, and high fat/high cholesterol. Cholesterol absorption values ranged widely from 36% to 74% on the low fat diet. Feeding a high fat or high fat/high cholesterol diet did not substantially change the percentage cholesterol absorption in these individuals.

Am J Physiol 1999; 276:G751-G760. This is a comprehensive study of the effects of cholic acid and cholesterol feeding in the mouse. Feeding cholesterol and cholic acid increased bile flow and biliary lipid secretion rates, and reduced hepatic bile acid synthesis. Intestinal bile acid absorption was dramatically increased. In contrast, feeding a diet containing cholesterol without bile acid did not produce these dramatic effects.

Marked reduction in bile acid synthesis in cholesterol 7x-hydroxylase-deficient mice does not lead to diminished tissue cholesterol turnover or to hypercholesterolemia. J Lipid Res 1998; 39:1833-1843. This study is a dramatic illustration of the importance of bile acids for intestinal cholesterol absorption and the multiple complex factors regulating cholesterol homeostasis. Despite a dramatic decrease for hepatic bile acid synthesis in the cholesterol 7x-hydroxylase knockout mice, total cholesterol turnover was actually increased and cholesterol balance across the animal was maintained.

Biochemistry 1998; 37:17843-17850. Indirect evidence is presented for a new candidate carrier that may mediate intestinal cholesterol absorption. The expression of SR-BI, an HDL receptor important for selective cholesteryl ester uptake in other tissues, was demonstrated in small intestine and Caco-2 cells. Cholesterol and cholesteryl ester was partially blocked in vitro by incubation with anti-SR-BI antibody.

Comparison of synthetic saponin cholesterol absorption inhibitors in rabbits: evidence for a non-stoichiometric, intestinal mechanism of action. J Lipid Res 1999; 40:464-474. The effects of a new generation saponin, pamaqueside, on cholesterol metabolism were evaluated. Rather than complex with cholesterol at a 1:1 ratio, this new saponin appeared to block cholesterol absorption at substoichiometric doses. This suggests a different mechanism of action, possibly by acting directly on the putative cholesterol transporter.

Ten well-characterized families with sitosterolemia were used for linkage analysis to localize the gene defect to chromosome 2p21. While there were no obvious candiate genes involved in lipid metabolism in this region, this is a critical first step toward identifying this gene that plays such an important role in plant sterol metabolism.

Intestinal absorption or dietary cholesteryl ester is decreased but retinyl ester absorption is normal in carboxyl ester lipase knockout mice. Biochemistry 1999; 38:4143-4149. Cholesterol and retinol absorption was examined in this knockout mouse. As reported previously, cholesterol absorption was unaffected and cholesteryl ester absorption was decreased by approximately 50%. In contrast to the cholesteryl ester, absorption of retinyl ester was unaffected in the CEL knockout mice. This indicated that another lipase besides CEL is important for retinol absorption.
