The molecular mechanism for the induction of hepatic sodium bile acid cotransport in postpartum rats was examined. These studies established that the induction is transcriptional and is mediated by prolactin acting through its receptor to facilitate Stat5 binding to interferon-γ-activated sequence-like elements in the NTCP promoter.

Gastroenterology 1997; 112:214-225. The effects of endotoxin administration on the transport of two bile acids (taurocholate and taurochenodeoxycholate) and two organic anions (BSP and taurolithocholate) were examined in the isolated perfused liver and isolated sinusoidal and canalicular membranes. The results showed that endotoxemia severely impaired transport of all four organic anions at the hepatic sinusoidal and canalicular membranes.

Intraduodenal infusion of taurocholate in biliary-diverted rats also induced transporter protein expression, indicating that the ASBT can be positively regulated by substrate.

Conditions were established for sustained expression of NTCP in a polarized hepatocyte cell system, WIF-B cells. This system overcomes many of the problems encountered with other hepatocyle cell lines and shows great promise for study of hepatocellular bile salt transport in vitro.

Am J Physiol 1997; 273:G842-G848. cAMP rapidly increases hepatocyte sodium-dependent bile acid uptake without requiring new transporter synthesis. These studies showed that cAMP increased NTCP protein content in plasma membranes without changing whole cell NTCP content. Thus, sodium-dependent bile acid transport can be regulated by plasma membrane transporter expression without changing hepatocyte NTCP expression.
