The molecular mechanism for the induction of hepatic sodium bile acid cotransport in postpartum rats was examined. These studies established that the induction is transcriptional and is mediated by prolactin acting through its receptor to facilitate Stat5 binding to interferon-γ-activated sequence-like elements in the NTCP promoter.

Gastroenterology 1997; 112:214-225. The effects of endotoxin administration on the transport of two bile acids (taurocholate and taurochenodeoxycholate) and two organic anions (BSP and taurolithocholate) were examined in the isolated perfused liver and isolated sinusoidal and canalicular membranes. The results showed that endotoxemia severely impaired transport of all four organic anions at the hepatic sinusoidal and canalicular membranes.

Interestingly, the elevated serum bile salt concentrations were not sufficient to down-regulate NTCP expression in these patients.
