    • Targeted deficiency or cytosolic truncation of the VE-cadherin gene in mice impairs VEGF-mediated endothelial survival and angiogenesis
    • ••] report on the latest knockout of a vertebrate classic cadherin. Mice that lack VE-cadherin die at mid-gestation showing multiple severe defects in vascular morphogenesis. VE-cadherin appears not to be essential for the formation of intercellular junctions between endothelial cells, as such junctions are still formed in mutant mice, although in smaller numbers and size. Carmeliet et al. also show that VE-cadherin is required for endothelial survival by forming a complex with β-catenin, the VEGF receptor-2, and phosphoinositide 3-kinase. This complex is essential for the reception of a survival signal mediated by the VEGF-A growth receptor.

Planar polarity in Drosophila epithelia is controlled by the frizzled pathway. Genetic interaction studies between dachsous and frizzled pathway genes suggests that Dachsous modulates the directionality of frizzled signaling. This finding represents the first evidence for the involvement of a cadherin in planar epithelial polarity.
