An important study of human humoral responses to hsp60 of C. trachomatist, showing for the first time an association between host antibody responses to hsp60 and trachomatous scarring. Subjects with trachomatous scarring had higher titers and a higher prevalence of antibodies against hsp60 compared with controls, who were subjects without clinical evidence of disease. This study also looks at the important relationship between genetic make up and immune response against hsp60.

This cell culture study has explored effect of IFN-γ on the growth of Chlamydia in human macrophages and has shown that infection of U937 cells was not affected by IFN-γ. There was also IFN-γ-independent persistence of atypical chlamydiae in these cells with the expression of MOMP, hsp60 and lipopolysaccharide.

An invaluable human study of conjunctival biopsies from subjects with trachoma and controls. The inflammatory cytokines and inflammatory cells were shown to be associated with trachoma. B lymphocytes were predominant compared with T lymphocytes in subjects with more superficial scarring, whereas T lymphocytes were predominant in subjects with more extensive scarring. Helper T cells outnumbered the cytotoxic T cells in all trachoma biopsies. Cytoplasmic IL-1α and β expression was seen in the conjunctival epithelium in all trachoma specimens. IL-1α and β, TNF-α and platelet-derived growth factor were detected in macrophages infiltrating the substantia propria. Trachoma subjects showed more inflammatory cells compared with controls and the expression of cytokines was absent in normal subjects.

B cell-deficient mice failed to produce an antibody response as well as a DTH response. The Th1 and Th2-related cytokines were also deficient after Chlamydia-specific in-vitro re-stimulation of the spleen cells from B cell knockout mice.

IL-6 knockout mice were used to study the role of this cytokine in murine lung tissue after intranasal infection with C, trachomatis, IL-6 knockout mice had significantly increased levels of chlamydia in the lung tissue and higher mortality rates compared with controls. The cytokine and antibody profile was compatible with a reduced but reversible Th1-like response in these mice. IL-6 therefore seems important in the early response against C. trachomatis infection.

The pre-exposed mice were also re-challenged 70 days later. Oral and subcutaneous infection produced the least response, whereas intranasal and intravenous infection produced the highest number of IFN-γ-producing T cells. Compared with intravenous infection, intranasal infection also produced comparable levels of IgA but higher levels of IgG2a in the vaginal secretions. Intranasally infected mice had the best protection on re-challenge.
