Saier MH, Paulsen JH, Sliwinski MK, Pao SS, Skurray RA, Nikaido H: Evolutionary origins of multidrug and drug-specific efflux pumps in bacteria. FASEB J 1998, 12:265-274. Analysis of complete genome sequences of five eubacterial and one archaebacterial species showed that putative drug efflux pumps occur widely. Major facilitator superfamily (MFS) pumps occur in large numbers (18 in E. coli) and very widely (found also in archaebacteria), but RND (resistance-nodulation-division) pumps are limited to Gram-negative eubacteria. Drug transporters occur in only 3 out of 17 subfamilies of MFS; in contrast, all of the RND pumps appear to be exporters of drugs, toxic metal ions, or lipooligosaccharides.

Nature 1998, 391:291-295. LmrA of L. lactis is a multidrug efflux pump homologous to one-half of the human multidrug transporter MDR1 or P-glycoprotein. This interesting study shows that when the ImrA gene is expressed in a human fibroblast line, the product not only goes to the plasma membrane, but also is fully functional in the extrusion of typical substrates of P-glycoprotein. Since expression of P-glycoprotein in bacteria is difficult, this system may serve as an ideal model system for future studies of P-glycoprotein.

The authors found that the blt gene, coding for an MFS (major facilitator superfamily) drug pump, is a part of an operon, apparently involved in detoxification of polyammes. This role was deduced because another gene of the operon codes for a polyamine acetylase. Indeed, spermidine is pumped out by cells overproducing Bit.

Nikaido H: The role of outer membrane and efflux pumps in the resistance of Gram-negative bacteria. Can we improve drug access? Drug Resistance Updates 1998,1:93-98. Poor access of many antibacterial agents to targets in Gram-negative bacteria is assumed to be caused by the synergy between the outer membrane barrier and the chromosomally coded multidrug efflux process. If so, we should be able to improve dramatically the efficacy of these drugs either by increasing the nonspecific permeability of the outer membrane, or by inhibiting the pump. This review examines this prediction critically, with experimental data.
