The purified protein catalyzes the synthesis of hexanoyl homoserine lactone (HL) from S-adenosylmethionine (SAM) and hexanoyl-acyl-ACP. The enzyme shows limited ability to generate acyl HLs when the hexanoyl group on the ACP is substituted with a butyryl or octanoyl moiety. This paper and reference [8••] are complementary.

The identification of a gene other than luxI that directs the synthesis of an acyl homoserine lactone (HL) in Vibrio fischeri is described. The product of this gene, AinS, shows no similarity to any member of the Luxl family. Rather, it is related to a protein implicated in acyl HL synthesis in V. harveyi. This suggests that these gene products represent a second, newly recognized family of autoinducer synthases.
