J Cell Biol 1996, 135:111-121 This paper, along with [15**] shows that cells lacking Pex13p, although deficient in import of matrix proteins retain membranes containing the peroxisomal membrane protein Pex11p. This observation indicates independent pathways for the integration of peroxisomal membrane proteins and for the translocation of peroxisomal matrix proteins. In addition, by screening databases of expressed sequence tags an open reading frame with the potential to encode an orthologue of the fungal Pex13p was identified in both the worm Caenorhabditis elegans and man. EMBO J 1997, 16:44-53. The authors, working with H. polymorpha, reported for the first time the identification of a PEX14 gene. It encodes a new peroxisomal membrane protein. Overproduction of Pex14p in wild-type H. polymorpha cells resulted in a peroxisome-deficient phenotype characterized by the presence of small vesicles which lacked matrix proteins. This result demonstrates that the stoichiometry of Pex14p and perhaps other peroxisomal membrane proteins is critical for matrix protein import.

Cell 1997, 89:83-92. Pex14p of Saccharomyces cerevisiae is shown to be a second component ot the peroxisomal translocation machinery. Most interestingly, based on the two-hybrid method and/or immunoprecipitation, it binds to four other components of the import machinery, the two membrane-bound peroxins, Pex13p and Pex17p, as well as the two peroxisomal targeting signal (PTS) receptors. Pex5p and Pex7p. These properties led to the proposal that Pex14p is the point of convergence of the PTS1- and PTS2-dependent import pathways, and provide strong evidence for the existence of a common translocation machinery for peroxisomal matrix proteins.

The peroxin Pex15p (383 amino acids), with a single transmembrane domain (332-349 amino acids), is an integral protein of the peroxisomal membrane in Saccharomyces cerevisiae. It was found to have surprising properties: it is the first peroxin shown to be phosphorylated and its carboxy-lerminal tail contains two overlapping sorting signals. Amino acids 302-353 target Pex15p to the endoplasmic reticulum (ER) where it becomes O-glycosylated. Amino acids 302-371 are required for targeting to the peroxisomes. These findings raise the possibility that the biogenesis of the peroxisomal membrane requires the involvement of the ER. At present, this is the best although still not completely convincing evidence that at least some peroxisomal membrane proteins are targeted to the peroxisome via the ER.

In this paper it is demonstrated that an interaction found with the yeast two-hybrid system can be mediated by a bridging molecule. By the use of gene deletions in the reporter strain PCY2 it is shown that Pex17p binds Pex14p but that it interacts with Pex5p indirectly through Pex14p.

Pex13p is an integral peroxisomal membrane protein with a cytoplasmic SH3 domain which interacts with Pex5p, the predominantly cytoplasmic PTS1 receptor.

As in [17**], it was shown to be a peroxisomal membrane protein and to bind also to Pex13p and to both PTS receptors, Pex5p and Pex7p, The possibility is discussed that Pex14p may be a component of the translocation machinery which functions either as a homo-oligomeric or hetero-oligomeric docking protein or as a releasing factor of the two cargo-loaded PTS receptors.

By studying the interesting effect of peroxisome-to-mitochondria mistargeting of the homodimeric enzyme alanine: glyoxylate aminotransferase1 (AGT), it is found that the autosomal recessive disease primary hyperoxaluria type 1 is associated with the combined presence of a normally occurring Prot 1 → Leu polymorphism and a disease-specific Gly170 → Arg mutation. While the former leads to the formation of a mitochondrial targeting sequence the combined presence of both mutations prevents homodimerization. Normal AGT rapidly dimerizes in the cytoplasm and is imported into peroxisomes as a dimer.

This paper reports results consistent with the notion that the import of peroxisomal matrix proteins is mediated by cycling receptors, as was previously suggested by Marzioch et al., 1994 [61]. It is nicely shown that human Pex5p, the predominantly cytoplasmic PTS1 receptor, accumulates at the peroxisomal membrane either when distinct peroxins (Pex2p and Pex12p) are lacking or when translocation of matrix proteins into peroxines is blocked. In the latter case, Pex5p returns to the cytoplasm when translocation is restored. Thus, it appears that the PTS receptor can cycle between cytoplasm and peroxisomes. Intriguingly, in PBD complementation group 7 Pex5p accumulates within the peroxisome. Whether the receptor actually enters the human peroxisome at one point of its cycle or whether this is the result of the altered activity of the peroxins deficient in these fibre-blasts is not yet known.
