Fluorescence experiments using a tetramethylrhodamine and fluorescein labeled duplex oligonucleotide were used to investigate the kinetics of binding and bending by yeast TBP. The time-dependent measurements obtained by stopped-flow fluorimetry showed that binding and bending of the DNA occur simultaneously and ended a long-standing debate.
