This study demonstrates the effect of staphylococcal enterotoxin (SE) A binding to the α and β chains of major histocompatibility complex (MHC) on recognition of the superantigen by specific Vβ elements. Mutations in SEA that affect its ability to interact with the low affinity class II α chain binding site resulted in stimulation of only two of the six Vβs that normally recognize the superantigen. By contrast, mutations that affect binding to the β chain of class II did not alter the Vβ recognition pattern. Together the data suggest that, for some superantigens, binding to the MHC α chain may be important to attain a specific orientation or conformation for recognition by certain Vβ subsets.

This study illustrates striking differences between staphylococcal enterotoxin (SE) A and SEE in their ability to mediate major histocompatibility complex (MHC)-independent T cells activation. Together with Malchiodi et al. 1995 [46], the study points to the fact that superantigens, including ones that are highly homologous to each other, have distinct modes of interaction with MHC and T-cell receptor.
