This study demonstrates the effect of staphylococcal enterotoxin (SE) A binding to the α and β chains of major histocompatibility complex (MHC) on recognition of the superantigen by specific Vβ elements. Mutations in SEA that affect its ability to interact with the low affinity class II α chain binding site resulted in stimulation of only two of the six Vβs that normally recognize the superantigen. By contrast, mutations that affect binding to the β chain of class II did not alter the Vβ recognition pattern. Together the data suggest that, for some superantigens, binding to the MHC α chain may be important to attain a specific orientation or conformation for recognition by certain Vβ subsets.

The results underscore the importance of superantigen-MHC interaction in regulating the magnitude of cytokine responses and controlling the clinical outcome of infection by superantigen-producing organisms.
