Characterization of protein kinase C β isoform activation on the gene expression of transforming growth factor-β, extracellular matrix components, and prostanoids in the glomeruli of diabetic rats. J Clin Invest 1996; 100:115-126. This is a very important paper, which expands on the data in Ref. 54. The authors show that in cultured mesangial cells glucose-induced biochemical abnormalities can be inhibited by LY333531. Furthermore, the treatment of diabetic rats with LY333531 inhibited increased mRNA for TGF-β1 and extracellular matrix proteins concomitant with the inhibition of elevated glomerular PKC activity.

Vascular endothelial growth-factor induced retinal permeability is mediated by protein kinase C in vivo and suppressed by an orally effective β-isoform selective inhibitor
-
Aiello LP, Bursell S-E, Clermont A, Duh E, Ishii H, Takagai C, et al. Vascular endothelial growth-factor induced retinal permeability is mediated by protein kinase C in vivo and suppressed by an orally effective β-isoform selective inhibitor. Diabetes 1997; 46:1473-1480. This is an important paper showing that although VEGF activates multiple PKCs in the retina, specific inhibition of PKCβ is sufficient to inhibit VEGF-induced changes in intraocular permeability.

Prevention of glomerular dysfunction in diabetic rats by treatment with d-α-tocopherol

Koya D, Lee I-K, Ishii H, Kanoh H, King GL. Prevention of glomerular dysfunction in diabetic rats by treatment with d-α-tocopherol. J Am Soc Nephrol 1997; 8:426-435. This paper shows that vitamin D prevents 1,2-diacylglycerol accumulation in the glomeruli of diabetic rats and normalizes renal function, which was associated with increased 1,2-diacylglycerol kinase activity, further supporting the importance of 1,2-diacylglycerol as a modulator of diabetic nephropathy by PKC activation.
