This study examined the CTL response of neonatal T cells to antigens presented by either nonprofessional or professional APCs. Neonatal T cells were equally responsive as adult T cells to antigens presented by professional APCs. On the other hand, neither neonatal or adult T cells responded to antigens presented by nonprofessional APCs. Therefore, the immune response of neonatal T cells are regulated by the functional properties of APCs, not by self-nonself discrimination.

    • Induction of TH1 and TH2 immunity in neonatal mice
    • Forsthuber T, Yip HC, Lehmann PV: Induction of TH1 and TH2 •• immunity in neonatal mice. Science 1996, 271:1728-1730. Neonatal T cells were immunized with antigens and their primary and secondary proliferative responses were examined and compared with the responses of adult T cells. Results showed that neonatal T cells were fully competent to secrete cytokines and help to generate antibody responses to antigen stimulation. As in adult T cells, the outcome of the neonatal T-cell responses was determined by the type of adjuvant. Antigens immunized with incomplete Freund's adjuvant induced a similar scale of Th2 responses while immunization with complete Freund's adjuvant resulted in Th1 responses in both neonates and adult mice. Thus, neonatal T cells were not specially susceptible to tolerance induction.

This is a recent review about the biological functions of CD28/CTLA-4 (CD152) and CD80/CD86 costimulatory systems. It gives detailed discussions of the putative roles of the costimulatory system in general T-cetl activation, transplantation, humoral responses and autoimmune responses. The differential effects of CD80 and CD86 were also thoroughly discussed. Finally, a 'strength-of-signal' model for CD28-CD80/CD86 costimulation was proposed to explain the differential effects.

J Immunol 1997,158:1572-1581. Non-depleting anti-CD4 mAb treatment blocked cardiac allograft rejection in LEW rats. Moreover, T cells isolated from these animals were able to transfer the graft tolerance to recipients. These T cells showed significantly diminished Th1 cytokine production but greatly enhanced Th2 cytokine production. Inhibition of Th2 cytokine production abrogated tolerance transfer capacities of these T cells.

GVDH blockade was associated with reduced production of IFN-γ, but the production of IL-4 and IL-10 was not affected.

This study showed that simultaneous blockade of CD80/CD86 and CD154 (CD40) signaling effectively abrogated T-cell responses and induced long-term acceptance of cardiac and skin allografts survival. Blocking CD80/CD86 or CD154 (CD40) alone only partially reduced T-cell responses and the allografts were ultimately rejected. These data indicate that tolerance is more likely be induced by the combined blockade of CD80/CD96 and CD154 (CD40).

Anti-CD86 mAb treatment blocked the first episode of EAE but was ineffective in preventing subsequent disease relapses. Surprisingly, combined treatment of anti-CD80 and anti-CD86 mAbs exacerbated the disease.

CD80 knockout mice showed similar antibody responses to wild-type mice, but CD86 knockout mice had more defects in antibody production when immunized without adjuvant. CD80/CD86 double knockout mice showed the most severe defects in their antibody responses.
