Cell 1995, 80:707-718. Mice treated with anti-B7-1 antibodies at the time of induction of EAE are protected from disease. This protection is due to immure deviation as shown by the cytokine phenotype of the immune response to the encephalitogenic peptide and by the cloning and transfer of protective, antigen-specific Th2 cells. Administration of anti-B7-2 antibodies exacerbated EAE and this was associated with an increase in levels of Th1 cytokines.

J Immunol 1995, 155:5011-5017. In this paper, the investigators developed techniques to generate polarized Th1 and Th2 populations in vitro. They generated the following populations: highly polarized Th1 cells which cause severe EAE on transfer into naive hosts and highly polarized Th2 cells which neither transfer disease nor confer protection on cotransfer with Th1 cells. They also found, however, that cells with a mixed phenotype, which caused mild disease on transfer into naive mice, diminished the severity of disease when cotransfered with Th1 cells.

This paper shows that such treatment induces a Th2-type cytokine response and that protection is partially reversed by the administration of a neutralizing anti-IL-4 antibody. Furthermore, treatment produced a striking reduction in the level of leukocyte infiltration within the brain.

Protection was associated with the production of anti-glutamate decarboxylase antibody isotypes consistent with a Th2 response, with a reduction in peptide-specific IFN-γ and an increase in peptide-specific IL-5. The data suggests that this treatment induced a regulatory Th2 response which inhibited the development of disease.

The phenotype of the immune response to the altered peptide is Th0/Th2 and therefore immune deviation has occurred despite immunization in complete Freund's adjuvant. T cells from mice immunized with the altered peptide can transfer protection to naive mice, and protection is partially reversed by treatment with anti-IL-4 monoclonal antibodies.

EAE induced by transfer of encephalitcgenic T cell lines or clones in the presence of a T cell clone specific for MBP87-99 was reversed by the administration of soluble native peptide or an analogue of it. The reversal of disease was prevented by neutralizing IL-4 and was associated with regression of inflammatory infiltrates within the brain.
