A strong correlation between mPPARγ activity and in vivo antidiabetic activity in mice was demonstrated; providing pharmacological evidence that PPARγ was the molecular target for the antidiabetic activity of these drugs.

J Biol Chem 1996, 271:29909-29914. The authors report that treatment of rats with the thiazolinedione BRL 49653 resulted in an increase in the mass of brown fat and increased expression of the uncoupling protein in this tissue. Cell culture studies showed that BRL 49653 regulated the expression of the uncoupling protein through activation of PPARγ.
