This paper provides confirmation of a role for ET-1 in the regulation of forearm vascular resistance and the first evidence that ET-1 plays a key role in regulation of systemic vascular resistance and blood pressure in humans. It also shows that endothelin receptor antagonists are capable of preventing 'vasospasm' to ET-1 in a forearm model.

    • Evidence for endothelin-1-mediated vasoconstriction in severe chronic heart failure
    • A/B receptor antagonist, bosentan, in patients with severe heart failure. The pattern of mixed reduction of pre- and after-load, without change in heart rate, is very favourable. There was no healthy control group with which to compare the haemodynamic effects, however, and the patients were studied after withdrawal from angiotensin-converting enzyme inhibitors.
