Elevated blood pressure and craniofacial abnormalities in mice deficient in endothelin-1

Nature 1994, 368:707-710. This paper is the first report on ET-1 null mice by gene targeting, showing an unexpected phenotype with craniofacial abnormalities, indicating that ET-1 is essential for the development of craniopharyngeal tissues. An interesting observation is that heterozygous mice presented with hypertension rather than hypotension.

It is interesting that defects in the ET-3 gene may cause Hirschsprung's disease in humans.

This paper describes the identification and characterization of ECE-1 from bovine adrenal glands. ECE-1 is a type II integral membrane-bound metalloprotease structurally homologous to neural endopeptidase 24,11 and Kell blood group protein. The proposed mechanism for its intracellular processing sites (Golgi complex and secretory vesicles), other than the plasma membrane, is intriguing.
