    • of outstanding interest. This study highlights the importance of En1 in D-V patterning and ridge formation. Targeted mutagenesis of En1 results in limbs in which ventral structures are transformed to a dorsal fate. This is correlated with ectopic expression of Wnt7a in the ventral ectoderm. Ridge formation is disrupted such that the ventral part of the ridge is expanded and flattened. Expression of the ridge markers Fgf8 and Bmp2 is also expanded.

    • Cyclopia and defective axial patterning in mice lacking Sonic hedgehog gene function
    • of outstanding interest. These studies unequivocally demonstrate the requirement for Shh signaling in development of the limb and other embryonic structures. Targeted disruptiong of Shh results in multiple embryonic structures. Loss of Shh function affects limb patterning and outgrowth presumably because of its roles in A-P patterning and in supporting P-D patterning via the positive feedback loop between Shh and Fgfs in the ridge.
