Ptoc Natl Acad Sci USA 1996, 93:4937-4941. The use of histidines to nucleate two-dimensional (2D) crystals extends the range of soluble proteins which may be crystallized in this manner. In addition to providing materials for structure determination by electron diffraction, 2D crystals can serve as seeds for exitaxial growth of three-dimensional crystals. Such crystals could also form the basic structures of biosensors.

Quenching of cooperative fluctuations and effects on protein stability. Biopolymers 1997, 41:131-143. This particular cross-link increases the unfolding temperature by 20°C. This raises the important issue of why so much. The authors suggest that the cross-link works by suppressing large-scale, concerted structural fluctuations.

    • Biochemistry and genetics of microbial xylanases
    • Jeffries TW: Biochemistry and genetics of microbial xylanases. Curr Opin Biotechnol 1996, 7:337-342. This paper provides a description of the major xylanase families, the important regions in substrate binding and catalysis, data on stability and pH dependence of activity, as well as some discussion of comercial applications. As such it provides a good starting point to gain a perspective on microbial xylanases.

    • Effects of both shortening and lengthening the active site nucleophile of Bacillus circulans xylanase on catalytic activity
    • Lawson SL, Wakarchuk WW, Withers SG: Effects of both shortening and lengthening the active site nucleophile of Bacillus circulans xylanase on catalytic activity. Biochemistry 1996, 35:10110-10118. The hydrolysis of glycosidase bonds can proceed by two mechanisms that are critically dependent upon the distance separating the carboxyl groups. In this work, the catalytic consequences of changing this distance are explored.
