The authors showed that there is a correlation between levels of mitochondrial function (as assessed by mitochondrial membrane potential) and the occurrence of either apoptotic or necrotic neuronal death following excitotoxicity: in this paradigm, necrotic deaths are seen more commonly in neurons whose mitochondria undergo extensive, irreversible loss of membrane potential.

In transgenic ALS mice, onset of motor neuron pathology, but not its rate of progression, was slowed by Vitamin E. The reciprocal situation was true for therapy with the anti-glutamate agent riluzole, suggesting that different biochemical events might underlie the initiation and the propagation of the motor neuron death process.
