Cells can become resistant to methotrexate through alteration of folate active transport mechanisms, but cells cannot acquire similar resistance to trimetrexate (TMTX) owing to the passive cellular entry of this drug. To overcome TMTX-induced myelosuppresssion, the L22Y DHFR mutant is used to provide increased TMTX resistance in transduced hematopoietic cells.
