49. 2 system, we also carried out a well-established religation assay with T4 ligase, but encountered problems when we tried to monitor the success or failure of the ligase reaction from previously isolated linear plasmid DNA. These problems might be due to ligase-DNA complex formation that hindered the movement of DNA in the agarose gel (also after phenol/chloroform extraction). As a control for the reliability of the assay, plasmid DNA cut by EcoRI and religated was used.
