Methods 1. Immunohistochemistry of TLR2,3,4,5 in Human Cornea Sections and Conjunctival Sections. Human corneal tissues were obtained from corneal buttons of a patient undergoing corneal transplantation for early-stage bullous keratopathy and human conjunctival tissues were obtained at conjunctivochalasis surgery. Serial sections (6 μm) were prepared and slides were fixed with methanol for 30 min. And then, slides were incubated with purified mouse anti-human TLR2 (TL2.1) or 3 (TLR3.7) or 4 (HTA125) monoclonal antibody (mAb; eBioscience, San Diego, CA) or TLR5 monoclonal antibody (mAb; Abcam, Cambridge, UK, or isotype control mouse IgG2a for TLR2,4,5, or mouse IgG1 for TLR3 (DakoCytomation, Kyoto, Japan) at 4 °C in a moist chamber overnight. After the slides were washed in PBS, Alexa Fluor 488 donkey anti-mouse IgG (H+L, Molecular Probes, Eugene, OR) was applied for 1 h at room temperature.
