    • Heterochromatin integrity affects chromosome reorganization after centromere dysfunction
    • Conditional deletion of an endogenous fission yeast centromere allows spontaneous assembly of a neocentromere at the subtelomere regions of the chromosome in addition to the intertelomere fusion to either of the remaining chromosomes. The balance between the two events changes dramatically by inactivating heterochromatin, suggesting a higher order regulation of chromosome reorganization by heterochromatin.

    • This study determines the copy numbers for the CENP-A and other structural proteins per kinetochore in C. albicans and S. pombe by using quantitative fluorescence microscopy. The calculated numbers of kinetochore protein complexes are essentially parallel to the number of microtubule attachment in both point and regional centromeres. But Cse4 in C. albicans appears to exist in excess, and yet the amount of both C. albicans Cse4 and S. pombe Cnp1 do not reflect the length of associating DNA regions.

    • ••] explore the essential role of the Scm3 protein in fission yeast. Unlike in S. cerevisiae, fission yeast Scm3 functions as a recruitment factor for Cnp1 at centromeres, and Scm3 itself dissociates from the centromeres during mitosis in conjunction with the Mis16-Mis18 complex [45]. In addition, both studies uncover a relative lack of histone H2A and H2B associated with the central domain of the centromere to which Cnp1 binds.

    • A NASP (N1/N2)-related protein, Sim3, binds CENP-A and is required for its deposition at fission yeast centromeres
    • This study explores a soluble nucleoplasmic protein Sim3, which interacts with Cnp1 and is required for deposition of Cnp1 at the centromere in S. pombe. Sim3 most probably escorts newly synthesized Cnp1 to the centromeres during S and G2 phases of the cell-cycle in a replication-independent manner.

    • Biphasic incorporation of centromeric histone CENP-A in fission yeast
    • Fission yeast Cnp1 deposition at the centromere occurs during S and G2 phases of the cell-cycle. The Cnp1 loading during S phase requires Ams2 protein, which is known to control histone gene expression. G2 phase loading pathway may compensate an inappropriate loading of Cnp1 in the adjacent S phase.

If the Cnp1 nucleosome is initially assembled on the plasmid, however, such kinetochore-forming chromatin will be maintained in the absence of pericentromeric heterochromatin.
