    • The authors describe the production of a leucine-zipper based stable galectin-1 homodimer (dGal) and examine its effect on human peripheral blood monocyte cells from healthy volunteers in vitro; they find that treatment with dGal attenuates IL-2 production and induces IL-10 and IL-1β production, suggesting that it may serve as a treatment for chronic inflammatory diseases.

    • The authors demonstrate that besides its role in enhancing proliferation of tumor-initiated cells, IL-6 produced by myeloid cells protects normal and premalignant intestinal epithelial cells from apoptosis; furthermore, they demonstrate that the effects of IL-6 are mediated by the transcription factor Stat3, whose ablation in intestinal epithelial cells portends a significant impact on tumorigenesis.

    • Identification and characterization of oligonucleotides that inhibit Toll-like receptor 2-associated immune responses
    • Using a novel molecular evolution strategy combining immunoprecipitation with the evolution of ligands by exponential enrichment (SELEX), the authors develop a method to facilitate the screening of high-affinity aptamers for TLR2; functional aptamers were then characterized using NF-κB reporter assays. One aptamer, AP177, significantly inhibits NF-κB activity, suppressing the secretion of cytokines by >80%, indicating therapeutic potential in the treatment of diseases characterized by a dysregulation of TLR2 signaling.

    • A novel Toll-like receptor 4 antagonist antibody ameliorates inflammation but impairs mucosal healing in murine colitis
    • The authors utilize two murine models of inflammatory bowel disease to determine the effects of TLR4/MD-2 antibody administration during induction or recovery from colitis; they find that Anti-TLR4 Ab treatment decreases macrophage and dendritic cell infiltrate and reduces IL-6 and TNF-α expression during initiation of colitis, but by contrast, treatment during recovery from colitis results in defective mucosal healing with decreased expression of COX-2, prostaglandin E2, and amphiregulin.
