A functional genomics approach reveals CHE as a component of the Arabidopsis circadian clock

The first published application of a high-throughput yeast-one-hybrid genomic strategy for the discovery of a plant transcription factor binding to a target promoter. To identify regulators of CCA1, the so-called 'promoter hiking' approach was used to screen a library of Arabidopsis transcription factors for binding to CCA1 promoter fragments. In a promoter hiking experiment, fragments of a promoter of interest are fused to the beta-galactosidase gene and inserted into yeast to produce bait strains. These bait strains are then systematically screened by transformation with a library consisting of hundreds of cloned transcription factors. Promoter hiking is effectively a high-throughput promoter bashing strategy that can theoretically be expanded to encompass all known and predicted transcription factors in an organism's proteome.
