In a promoter hiking experiment, fragments of a promoter of interest are fused to the beta-galactosidase gene and inserted into yeast to produce bait strains. These bait strains are then systematically screened by transformation with a library consisting of hundreds of cloned transcription factors. Promoter hiking is effectively a high-throughput promoter bashing strategy that can theoretically be expanded to encompass all known and predicted transcription factors in an organism's proteome.
