Using specific IL-13/IL-4 inhibitor prior to sensitization inhibited development of the allergic phenotype. Once allergy was established, however, this mode of inhibition was not effective. This study suggests that mechanisms of allergic primary and secondary responses are distinct.

Two models of allergic airway disease were compared in this study examining the effects of diesel exhaust and ambient particles. The authors noted differences in allergic responses depending upon the animal model used and showed pollutant particles can act as adjuvants, increasing levels of allergen-specific IgE and inflammation in a mouse model of allergic airways disease.

Tumor necrosis factor-α knockout mice had reduction in levels of ovalbumin-specific IgE, decreased inflammation and symptom scores when compared with wild-type. The authors suggest that tumor necrosis factor-α may be important for stimulation of adhesion molecules important for recruitment of inflammatory cells.

CD4+ T cells expressing IL-10 specifically inhibited airway hyperreactivity and eosinophilia in both BALB/c and reconstituted severe combined immune deficiency mice. This effect was neutralized by anti-IL-10 antibody. These data support a important regulatory role for IL-10 cytokine.
