The Hsieh-Wilson laboratory used a chemoenzymatic strategy in which they probed O-GlcNAc-glycosylated proteins from rat forebrain extract with a ketone-biotin tag. By combining this isolation strategy with BEMAD, they were also able to identify the sites of glycosylation.
