A common ankyrin-G-based mechanism retains KCNQ and Nav channels at electrically active domains of the axon

The authors demonstrate that the targeting of KCNQ2/3 Kv channels to axon initial segments (and probably nodes of Ranvier) depends on binding to ankyrinG. KCNQ2/3 channels contain a motif nearly identical to that found in Nav channels that also mediates its interaction with ankyrinG. Finally, using comparative genomics, the authors propose this motif represents an example of convergent molecular evolution in Nav and Kv channels at about the time myelin evolved.

Using a full knockout of the neurofascin gene (Nfasc-null), the authors clearly demonstrate that NF-186 is necessary for the clustering of Nav channels and other proteins associated with nodes of Ranvier. They also prove that NF-155 is necessary for paranodal junction formation as the paranodal defect in the Nfasc-null mouse can be rescued by introducing an epitope-tagged NF-155 into myelinating glia.

Does paranode formation and maintenance require partitioning of neurofascin 155 into lipid rafts?

This study suggests that the assembly of paranodal junctions depends on the formation of paranodal lipid rafts. The localization of NF-155 to paranodal junctions was shown to be co-incident with its partitioning into biochemically distinct, lipid-rich membrane microdomains. The authors propose that disruption of these membrane microdomains explains the defects observed in paranodal mutant animals lacking myelin galactolipids.

Spectrins and AnkyrinB constitute a specialized paranodal cytoskeleton

This paper reports the identification of a specialized paranodal cytoskeleton comprised of ankyrinB, αII spectrin and βII spectrin. Furthermore, the authors show, using Caspr-null paranodal mutants, that organization of the paranodal cytoskeleton depends on neuron-glia interactions.

Disruption of axo-glial junctions causes cytoskeletal disorganization and degeneration of Purkinje neuron axons

This study addresses the role of paranodal axoglial junctions in the maintenance of the axonal cytoskeleton and axonal integrity. The authors show that disruption of paranodal junctions, as occurs in Caspr-null and CGT-null mice, results in a disorganized cytoskeleton and axon degeneration. These data could offer insight into underlying mechanisms of neurodegenerative disorders.
