    • In vitro enzymatic activity of FAS was determined by monitoring the production of free coenzyme A (CoA-SH) from malonyl- and acetyl-CoA during the FAS enzymatic reaction. CoA-SH levels were determined using the thiol-reactive dye 7-diethylamino-3-(4-maleimidophenyl)-4-methylcoumarin (CPM). Unreacted CPM is nonfluorescent, whereas the CPM-CoA adduct shows a peak fluorescence emission at 475 nM after excitation at 380 nM.
