    • Dectin-1 mediates macrophage recognition of Candida albicans yeast but not filaments
    • During the course of mammalian infection the transition of C. albicans from yeast to the filamentous state is crucial for pathogenicity. This study demonstrated that yeast cells accumulate surface β-glucans through bud and birth scars, whereas hyphae did not. In contrast to yeast cells, hyphae failed to activate dectin-1-dependent signaling, leading to diminished macrophage antifungal effector functions.

    • Aspergillus fumigatus triggers inflammatory responses by stage-specific beta-glucan display
    • •] revealed that A. fumigatus conidia provoke inflammatory responses only if fungal cells could undergo swelling, a process that led to β-glucan exposure. TLR/MyD88 and dectin-1 signaling were both activated by conidial swelling but acted independently to produce inflammatory mediators. Selective innate immune recognition of germinating conidia might provide a mechanism to limit potentially harmful inflammatory responses in the lung to conidia that represent an invasive threat.

    • The authors designed a vaccine that generates β-glucan-specific antibodies that can provide immunity to Candida and Aspergillus infections in preclinical animal studies without the need for cell-mediated killing mechanisms. The promise of this approach is the induction or infusion of anti-β-glucan antibodies that have antifungal effector properties for prophylactic benefit in vulnerable hosts or for therapeutic benefit in infected hosts.

    • The authors investigated the protective effect of myeloid-committed progenitor cells to mice infected with A. fumigatus during chemotherapy-induced neutropenia. This preclinical study demonstrated that progenitors entered the bone marrow, differentiated into neutrophils, and migrated to sites of infection, which resulted in enhanced survival.
