Gundersen Nuclear movement regulated by cdc42, MRCK, myosin, and actin flow establishes MTOC polarization in migrating cells Cell 121 2005 451 463 Here, the authors examine the mechanism of MTOC reorientation during cell migration and make two important findings. Firstly, during MTOC reorientation, it is not the MTOC that moves per se; rather, the nucleus moves in a retrograde fashion to position itself behind the MTOC. Secondly, Par6 functions to 'hold' the MTOC in a fixed position while the nucleus is moving.

This was one of the first genetic studies to elucidate a hierarchy and sequence of functioning among Cdc42, Par6 and aPKC. Furthermore, this paper demonstrated that the antagonism between the Par6 polarity complex and Lgl is actually mutual.

They discovered that the polarity complex regulates the interaction between APC and Dlg. This finding may underlie the mechanism by which MTOC reorientation occurs in relation to extracellular cues.
