Depletion of WSTF by RNA interference results in the assembly of an aberrant, condensed chromatin structure, enriched with markers of heterochromatin such as HP1.

Yeast RSC complex subunits interact directly with cohesin subunits. Impairment of the RSC complex containing Rsc2p prevents the cohesin complex from associating with chromosome arms, but still allows binding to centromeres. This suggests a role for this RSC complex in loading cohesin onto chromosome arms specifically, which in turn is required for sister chromatid pairing and chromosome segregation.

The architectural transcription factor NHP6A (non-histone protein 6) is a multi-copy suppressor of arp7 and arp9 conditional alleles. RSC physically interacts with Nhp6a and facilitates its binding to nucleosomes in vitro. This work suggests that ARP heterodimers work with architectural proteins of the HMG type to regulate chromatin architecture.
