Using cell-specific gene targeting, the allele dose of VEGF was altered ONLY in the podocyte. Each VEGF genotype was associated with a dramatic and distinct glomerular phenotype demonstrating an exquisite dosage-sensitivity for VEGF within the developing glomerulus. The glomerular phenotypes resembled congenital nephropathy, endotheliosis of preeclampsia and HIV-associated nephropathy.

Identified elevated levels of sFlt-1 in patients with preeclampsia and showed that injection of rats with an adenoviral vector encoding sFlt-1 led to endotheliosis, hypertension and proteinuria, the clinical signs of preeclampsia. Together, their results provide a plausible hypothesis for organ dysfunction in preeclampsia that can be tested prospectively in patients.
