Rotello Monolayer-controlled substrate selectivity using noncovalent enzyme-nanoparticle conjugates J Am Chem Soc 126 2004 13572 13573 This study demonstrates the control of substrate specificity by the surface in a surface-protein interaction. The enzymatic activity of monolayer-bound protein was examined with cationic, anionic and neutral substrates, revealing that the chemoselectivity of substrates originated from a combination of steric hindrance and electrostatic interactions. This control over enzymatic behavior is essential for fabrication of bioactive surfaces and materials.

Compared with commonly used metal-chelate affinity chromatography, the pre-equilibration step is obviated in the magnetic nanoparticle system. In addition, the recombinant proteins can be conveniently transferred to a desired substrate and anchored onto it by an external magnet.

This bio-bar-code method is superior to the current protein detection methods in many aspects, which include high sensitivity, prompt assay and low background. Moreover, it has the potential for massive multiplexing and the simultaneous detection of many analytes in one solution.
