The permeabilities and plate heights for these columns were compared with Chromolith columns, and separation impedances as low as 200 were measured. The authors speculate that performance limitations exist for silica monoliths at very high flow velocities.

Results are rationalized using a dimensionless ratio of two characteristic lengths, one characterizing the pressure drop and the other characterizing band broadening.

It is shown that increases in packing heterogeneity yield large increases in plate height and separation impedance.

Among other results, it is concluded that much lower plate heights can be achieved in currently available silica monoliths by making the structure more uniform, as opposed to making the size of the through-pores smaller.

After mechanical separation of the fusion protein attached to the PHB granules, the fusion protein can be self-cleaved to release the target protein in a highly purified form.
