Unlike some other isolates, E. muris causes a nonfatal persistent infection in the mouse. Bacteria persisted for at least as long as 5 months post infection in the spleen, liver and lung. Liver pathology was relatively mild, and hepatic necrosis was largely absent. The mouse model provides an opportunity to study how the bacteria evade host immune responses over a prolonged period.

Other data suggest that memory responses elicited by IOE may be impaired.

Prior infection with E. muris was sufficient to provide protection against fatal IOE challenge. Other evidence suggested that inflammatory cytokine production by CD8 T cells during fatal IOE challenge contributed to immunopathology.

Evidence is provided that E. chaffeensis modulates innate receptor signaling in response to lipopolysaccharide by downregulating TLR2 and 4 and CD14 during infection of human monocytes. Downregulation may be achieved via inhibition of p38 MAPK and Erk 1/2 activities.

A related rickettsia, A. phagocytophila, was able to establish infection in endothelial cell lines, suggesting that endothelial cells may be targets of rickettsia infection in vivo. During tick infection bacteria do not have direct access to blood vessels, so endothelial cell infection may allow eventual dissemination into the blood stream.
